On 29th March 1519 [his father] Francesco Gonzaga II Marquess of Mantua [aged 52] died. Federico Gonzaga II Duke Mantua [aged 18] succeeded II Marquess Mantua.

On 25th March 1530 Federico Gonzaga II Duke Mantua [aged 29] was created II Duke Mantua.

On 10th March 1533 [his son] Francesco Gonzaga III Duke Mantua was born to Federico Gonzaga II Duke Mantua [aged 32] and Margaret Palaeologina Duchess Mantua [aged 22]. He married 22nd October 1549 his fifth cousin Catherine of Austria Queen Consort Poland, daughter of Ferdinand I Holy Roman Emperor and Anne Jagiellon.

On 24th April 1538 [his son] William Gonzaga I Duke Mantua was born to Federico Gonzaga II Duke Mantua [aged 37] and Margaret Palaeologina Duchess Mantua [aged 27]. He married his fifth cousin Eleanor of Austria Duchess Mantua, daughter of Ferdinand I Holy Roman Emperor and Anne Jagiellon, and had issue.

On 13th February 1539 [his mother] Isabella Este [aged 64] died.
